Usage Notes
* Do not mix components from different kit lots or use reagents beyond the kit expiration date.
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Preparation and Storage
Store at 2-8 degree C.

Description: This Human Twinfilin-1 (TWF1) ELISA Kit employs a two-site sandwich ELISA to quantitate TWF1 in samples. An antibody specific for TWF1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any TWF1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TWF1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TWF1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: TWF1 encodes twinfilin, an actin monomer-binding protein conserved from yeast to mammals. Studies of the mouse counterpart suggest that this protein may be an actin monomer-binding protein, and its localization to cortical G-actin-rich structures may be regulated by the small GTPase RAC1. The deduced 350-amino acid protein has a calculated molecular mass of 40.3 kD. TWF1 has an N-myristoylation site and several sites for threonine, serine, and tyrosine phosphorylation, but it lacks a conserved protein kinase catalytic domain. Northern blot analysis detected a 3.4-kb transcript that was highly expressed in colon, testis, uterus, ovary, prostate, and lung. Lower expression was detected in brain, bladder, and heart, and no expression was detected in liver.

By capping the barbed ends of filaments, it also regulates motility. Seems to play an important role in clathrin-mediated endocytosis and distribution of endocytic organelles. Interacts with G-actin; ADP-actin form and capping protein (CP). May also be able to interact with TWF2 and phosphoinositides, PI(4,5)P2. When bound to PI(4,5)P2, it is down- regulated. Expressed at high levels in the colon, testis, ovary, prostate and lung. Expressed at lower levels in the brain, bladder and heart. Not detected in liver. Belongs to the actin-binding proteins ADF family. Twinfilin subfamily. 3 isoforms of the human protein are produced by alternative splicing.
