Usage Notes
* Do not mix components from different kit lots or use reagents beyond the kit expiration date.
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Preparation and Storage
Store at 2-8 degree C.

Description: This Human Tumor suppressor p53-binding protein 1 (TP53BP1) ELISA Kit employs a two-site sandwich ELISA to quantitate TP53BP1 in samples. An antibody specific for TP53BP1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any TP53BP1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TP53BP1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TP53BP1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: TP53BP1 has a predicted molecular mass of 217 kD and shows homology in its C-terminal 247 amino acids to the C terminus of the tumor suppressor protein BRCA1. TP53BP1 was present in both the cytoplasm and nucleus in some cells and only in the nucleus in others. Furthermore, there were 2 nuclear staining patterns, one homogeneous staining and the other dot staining.TP53BP1 was required for p53 accumulation, G2/M checkpoint arrest, and the intra-S-phase checkpoint in response to ionizing radiation. TP53BP1 played a partially redundant role in phosphorylation of the downstream checkpoint effector proteins BRCA1 and CHK2 but was required for the formation of BRCA1 foci in a hierarchical branched pathway for the recruitment of repair and signaling proteins to sites of DNA damage.

NCBI Description

This gene encodes a protein that functions in the DNA double-strand break repair pathway choice, promoting non-homologous end joining (NHEJ) pathways, and limiting homologous recombination. This protein plays multiple roles in the DNA damage response, including promoting checkpoint signaling following DNA damage, acting as a scaffold for recruitment of DNA damage response proteins to damaged chromatin, and promoting NHEJ pathways by limiting end resection following a double-strand break. These roles are also important during V(D)J recombination, class switch recombination and at unprotected telomeres. Alternative splicing results in multiple transcript variants encoding different isoforms. [provided by RefSeq, Aug 2017]

53BP1: a DNA damage checkpoint protein that interacts with HDAC4 protein to mediate the DNA damage response. Inhibits E2F1-mediated apoptosis in prostate cancer cells. Recruited to double strand breaks via the binding of its Tudor domain to dimethylated H3 K20. Asymmetrically dimethylated on Arg residues by PRMT1. Methylation is required for DNA binding. Binds to the central domain of p53. Interacts with Artemis. Interacts with histone H2AFX and this requires phosphorylation of H2AFX on S139. Interacts with histone H4 that has been dimethylated at K20. Has low affinity for histone H4 containing monomethylated K20. Does not bind histone H4 containing unmethylated or trimethylated K20. Has low affinity for histone H3 that has been dimethylated on K79. Does not bind unmethylated histone H3. Phosphorylated at basal level in the absence of DNA damage. Hyper-phosphorylated in an ATM-dependent manner in response to DNA damage induced by ionizing radiation. Hyper-phosphorylated in an ATR-dependent manner in response to DNA damage induced by UV irradiation. Two alternatively spliced human isoforms have been observed.
