Intended Uses: This kit is a sandwich enzyme immunoassay for in vitro quantitative measurement of TNFRSF8 in human serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids.

Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to TNFRSF8. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody preparation specific to TNFRSF8. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain TNFRSF8, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of TNFRSF8 in the samples is then determined by comparing the O.D. of the samples to the standard curve.

The protein encoded by this gene is a member of the TNF-receptor superfamily. This receptor is expressed by activated, but not by resting, T and B cells. TRAF2 and TRAF5 can interact with this receptor, and mediate the signal transduction that leads to the activation of NF-kappaB. This receptor is a positive regulator of apoptosis, and also has been shown to limit the proliferative potential of autoreactive CD8 effector T cells and protect the body against autoimmunity.
