Intended Uses: This ELISA kit can be applied to the in vitro quantitative determination of Human TFR/CD71 concentrations in serum, plasma and other biological fluids.

Principle of the Assay: This ELISA kit uses Sandwich-ELISA as the method. The micro ELISA plate provided in this kit has been precoated with an antibody specific to Human TFR/CD71. Standards or samples are added to appropriate micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibodies specific for Human TFR/CD71 and Avidin-Horseradish Peroxidase (HRP) conjugate are added to each micro plate well successively and incubated. After incubation, free components are washed away. Then the Substrate Reagent is added to each well, only those wells that contain Human TFR/CD71, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction will be terminated by adding Stop Solution and appears yellow in color. The optical density (OD) can be measured with spectrophotometry at a wavelength of 450 nm ± 2 nm. The OD value is proportional to the concentration of Human TFR/CD71. The concentration of Human TFR/CD71 in samples can be calculated by comparing the OD of the samples with the standard curve.

This gene encodes a cell surface receptor necessary for cellular iron uptake by the process of receptor-mediated endocytosis. This receptor is required for erythropoiesis and neurologic development. Multiple alternatively spliced variants have been identified. [provided by RefSeq, Sep 2015]

TfR: the transferrin receptor. Regulates the cellular uptake of iron occurs via receptor-mediated endocytosis of ligand-occupied receptors into specialized endosomes. Endosomal acidification leads to iron release. The apotransferrin-receptor complex is then recycled to the cell surface with a return to neutral pH and the concomitant loss of affinity of apotransferrin for its receptor. Transferrin receptor is necessary for development of erythrocytes and the nervous system.
