The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Teratocarcinoma Derived Growth Factor 1 (TDGF1). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Teratocarcinoma Derived Growth Factor 1 (TDGF1). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Teratocarcinoma Derived Growth Factor 1 (TDGF1), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Teratocarcinoma Derived Growth Factor 1 (TDGF1) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

This gene encodes an epidermal growth factor-related protein that contains a cripto, FRL-1, and cryptic domain. The encoded protein is an extracellular, membrane-bound signaling protein that plays an essential role in embryonic development and tumor growth. Mutations in this gene are associated with forebrain defects. Pseudogenes of this gene are found on chromosomes 2, 3, 6, 8, 19 and X. Alternate splicing results in multiple transcript variants. Could play a role in the determination of the epiblastic cells that subsequently give rise to the mesoderm (PubMed:11909953).
