Usage Notes
* Do not mix components from different kit lots or use reagents beyond the kit expiration date.
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Preparation and Storage
Store at 2-8 degree C.

Related Product Information for TMF1 elisa kit
Description: This Human TATA element modulatory factor (TMF1) ELISA Kit employs a two-site sandwich ELISA to quantitate TMF1 in samples. An antibody specific for TMF1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any TMF1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TMF1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TMF1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: The TATA element, or TATA box, is a critical cis-acting regulatory element in many promoters transcribed by RNA polymerase II. The RNA polymerase II transcription factor D is a biochemically defined HeLa cell nuclear fraction containing a transcription factor activity that binds specifically to the TATA box. The DNA-binding subunit of TFIID is the TATA box-binding protein. TMF binds to the HIV-1 TATA element in gel-retardation assays and inhibits activation of the HIV-1 long terminal repeat by TBP in in vitro transcription assays. Sequence analysis of the TMF cDNA revealed leucine zipper motifs and homology in its DNA-binding domain with the phage-encoded DNA binding protein Ner. In cataloging sequences from a human adult testis expression library, Affara et al. (1994) also identified a cDNA clone for TMF.

Uniprot Description

TMF1: Potential coactivator of the androgen receptor. Mediates STAT3 degradation. May play critical roles in two RAB6-dependent retrograde transport processes: one from endosomes to the Golgi and the other from the Golgi to the ER. This protein binds the HIV-1 TATA element and inhibits transcriptional activation by the TATA-binding protein (TBP). 2 isoforms of the human protein are produced by alternative splicing.

Protein type: Transcription factor; Nuclear receptor co-regulator

Chromosomal Location of Human Ortholog: 3p14.1

Cellular Component: Golgi membrane; Golgi apparatus; endoplasmic reticulum; nucleus

Molecular Function: protein binding; DNA binding; transcription cofactor activity

Biological Process: acrosome formation; transcription from RNA polymerase II promoter; positive regulation of cytokine production; luteinizing hormone secretion; sperm motility; Leydig cell differentiation; regulation of transcription, DNA-dependent; defense response to bacterium; spermatid nuclear differentiation; negative regulation of apoptosis

Product Notes

The Human TMF1 tmf1 (Catalog #AAA9713928) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA9713928 ELISA Kit recognizes Human TMF1. It is sometimes possible for the material contained within the vial of "TATA Element Modulatory Factor (TMF1), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
