Principle of the Assay: SYN-1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-SYN-1 antibody and an SYN-1-HRP conjugate. The assay sample and buffer are incubated together with SYN-1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the SYN-1 concentration since SYN-1 from samples and SYN-1-HRP conjugate compete for the anti-SYN-1 antibody binding site. Since the number of sites is limited, as more sites are occupied by SYN-1 from the sample, fewer sites are left to bind SYN-1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The SYN-1 concentration in each sample is interpolated from this standard curve.
NCBI and Uniprot Product Information
NCBI Description
Many different human endogenous retrovirus (HERV) families are expressed in normal placental tissue at high levels, suggesting that HERVs are functionally important in reproduction. This gene is part of an HERV provirus on chromosome 7 that has inactivating mutations in the gag and pol genes. This gene is the envelope glycoprotein gene which appears to have been selectively preserved. The gene's protein product is expressed in the placental syncytiotrophoblast and is involved in fusion of the cytotrophoblast cells to form the syncytial layer of the placenta. The protein has the characteristics of a typical retroviral envelope protein, including a furin cleavage site that separates the surface (SU) and transmembrane (TM) proteins which form a heterodimer. Alternatively spliced transcript variants encoding the same protein have been found for this gene. [provided by RefSeq, Mar 2010]
Uniprot Description
This endogenous retroviral envelope protein has retained its original fusogenic properties and participates in trophoblast fusion and the formation of a syncytium during placenta morphogenesis. May induce fusion through binding of SLC1A4 and SLC1A5 (PubMed:10708449, PubMed:12050356, PubMed:23492904).

Product Notes
The Human ERVW-1 ervw-1 (Catalog #AAA7254495) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA7254495 ELISA Kit recognizes Human ERVW-1. It is sometimes possible for the material contained within the vial of "Syncytin 1, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
