Human Repulsive guidance molecule A (RGMA) ELISA Kit
Principle of the Assay: RGMA ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-RGMA antibody and an RGMA-HRP conjugate. The assay sample and buffer are incubated together with RGMA-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the RGMA concentration since RGMA from samples and RGMA-HRP conjugate compete for the anti-RGMA antibody binding site. Since the number of sites is limited, as more sites are occupied by RGMA from the sample, fewer sites are left to bind RGMA-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The RGMA concentration in each sample is interpolated from this standard curve.

The encoded protein is a glycosylphosphatidylinositol-anchored glycoprotein that functions as an axon guidance protein in the developing and adult central nervous system. This protein may also function as a tumor suppressor in some cancers. Alternate splicing results in multiple transcript variants. [provided by RefSeq, Oct 2009]

Regulates cephalic neural tube closure, inhibits neurite outgrowth and cortical neuron branching, and the formation of mature synapses. Binding to its receptor NEO1/neogenin induces activation of RHOA-ROCK1/Rho-kinase signaling pathway through UNC5B-ARHGEF12/LARG-PTK2/FAK1 cascade, leading to collapse of the neuronal growth cone and neurite outgrowth inhibition. Furthermore, RGMA binding to NEO1/neogenin leads to HRAS inactivation by influencing HRAS1-PTK2/FAK1-AKT1 pathway. It also functions as a bone morphogenetic protein (BMP) coreceptor that may signal through SMAD1, SMAD5, and SMAD8. Belongs to the repulsive guidance molecule (RGM) family. 2 isoforms of the human protein are produced by alternative splicing.
