Intended Uses: This RELN ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of HumanRELN. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: RELN ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-RELN antibody and an RELN-HRP conjugate. The assay sample and buffer are incubated together with RELN-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the RELN concentration since RELN from samples and RELN-HRP conjugate compete for the anti-RELN antibody binding site. Since the number of sites is limited, as more sites are occupied by RELN from the sample, fewer sites are left to bind RELN-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The RELN concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a large secreted extracellular matrix protein thought to control cell-cell interactions critical for cell positioning and neuronal migration during brain development. This protein may be involved in schizophrenia, autism, bipolar disorder, major depression and in migration defects associated with temporal lobe epilepsy. Mutations of this gene are associated with autosomal recessive lissencephaly with cerebellar hypoplasia. Two transcript variants encoding distinct isoforms have been identified for this gene. reelin: Extracellular matrix serine protease that plays a role in layering of neurons in the cerebral cortex and cerebellum. Regulates microtubule function in neurons and neuronal migration. Affects migration of sympathetic preganglionic neurons in the spinal cord, where it seems to act as a barrier to neuronal migration. Enzymatic activity is important for the modulation of cell adhesion. Binding to the extracellular domains of lipoprotein receptors VLDLR and LRP8/APOER2 induces tyrosine phosphorylation of DAB1 and modulation of TAU phosphorylation. Defects in RELN are the cause of lissencephaly type 2 (LIS2); also known as lissencephaly with cerebellar hypoplasia or Norman-Roberts syndrome. LIS2 is a classic type lissencephaly associated with abnormalities of the cerebellum, hippocampus and brainstem. Individuals with LIS2 are severely ataxic, mentally retarded and suffer from epilepsy. Belongs to the reelin family. 3 isoforms of the human protein are produced by alternative splicing.
