Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti-OPN polyclonal antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-OPN polyclonal antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the OPN amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of OPN can be calculated!!Background/Introduction: Osteopontin (OPN), also known as bone sialoprotein I (BSP-1 or BNSP), secreted phosphoprotein 1 (SPP1), is a human gene product. The gene has 7 exons, spans 5 kilobases in length and in humans it is located on 4q13. The protein is composed of ~300 amino acids residues and has ~30 carbohydrate residues attached including 10 sialic acid residues, which are attached to the protein during post-translational modification in the Golgi apparatus. Osteopontin has been implicated as an important factor in bone remodeling. Specifically, research suggests it plays a role in anchoring osteoclasts to the mineral matrix of bones.

NCBI Description

This gene is one of several chemokine genes clustered on the q-arm of chromosome 17. Chemokines form a superfamily of secreted proteins involved in immunoregulatory and inflammatory processes. The superfamily is divided into four subfamilies based on the arrangement of the N-terminal cysteine residues of the mature peptide. This chemokine, a member of the CC subfamily, functions as a chemoattractant for blood monocytes, memory T helper cells and eosinophils. It causes the release of histamine from basophils and activates eosinophils. This cytokine is one of the major HIV-suppressive factors produced by CD8+ cells. It functions as one of the natural ligands for the chemokine receptor chemokine (C-C motif) receptor 5 (CCR5), and it suppresses in vitro replication of the R5 strains of HIV-1, which use CCR5 as a coreceptor. Alternative splicing results in multiple transcript variants that encode different isoforms. [provided by RefSeq, Jul 2013]

Uniprot Description

Function: Chemoattractant for blood monocytes, memory T-helper cells and eosinophils. Causes the release of histamine from basophils and activates eosinophils. Binds to CCR1, CCR3, CCR4 and CCR5. One of the major HIV-suppressive factors produced by CD8+ T-cells. Recombinant RANTES protein induces a dose-dependent inhibition of different strains of HIV-1, HIV-2, and simian immunodeficiency virus (SIV). The processed form RANTES(3-68) acts as a natural chemotaxis inhibitor and is a more potent inhibitor of HIV-1-infection. The second processed form RANTES(4-68) exhibits reduced chemotactic and HIV-suppressive activity compared with RANTES(1-68) and RANTES(3-68) and is generated by an unidentified enzyme associated with monocytes and neutrophils. Ref.3 Ref.8 Ref.10 Ref.11 Ref.12

Subcellular location: Secreted.

Tissue specificity: T-cell and macrophage specific.

Induction: By mitogens.

Post-translational modification: N-terminal processed form RANTES(3-68) is produced by proteolytic cleavage, probably by DPP4, after secretion from peripheral blood leukocytes and cultured sarcoma cells. The identity of the O-linked saccharides at Ser-27 and Ser-28 are not reported in Ref.8. They are assigned by similarity.

Polymorphism: The variant Phe-24 is an antagonist of the chemokine receptors CCR1 and CCR3.

Sequence similarities: Belongs to the intercrine beta (chemokine CC) family.

Mass spectrometry: Molecular mass is 7515±1 Da from positions 27 - 91. Determined by SELDI. Ref.12Molecular mass is 7862.8±1.1 Da from positions 24 - 91. Determined by ESI. Ref.8Molecular mass is 8355±10 Da from positions 24 - 91. Determined by ESI. O-glycosylated. Ref.8

Product Notes

The Human RANTES ccl5 (Catalog #AAA355338) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA355338 ELISA Kit recognizes Human RANTES. AAA Biotech's RANTES can be used in a range of immunoassay formats including, but not limited to, Quantitative sELISA (EIA). For quantitative detection of Rantes in human serum, plasma, body fluids, tissue lysates or cell culture supernatants. Researchers should empirically determine the suitability of the RANTES ccl5 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "RANTES, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
