Principle of the Assay: The Picokine™ Human QPCT Pre-Coated ELISA (Enzyme-Linked Immunosorbent Assay) kit is a solid phase immunoassay specially designed to measure Human QPCT with a 96-well strip plate that is pre-coated with antibody specific for QPCT. The detection antibody is a biotinylated antibody specific for QPCT. The capture antibody is a monoclonal antibody from mouse, the detection antibody is a biotinylated monoclonal antibody from goat. The kit contains recombinant Human QPCT with immunogen: Expression system for standard: NSO, Immunogen sequence: V29-L361. The kit is analytically validated with ready to use reagents. To measure Human QPCT, add standards and samples to the wells, then add the biotinylated detection antibody. Wash the wells with PBS or TBS buffer, and add Avidin-Biotin-Peroxidase Complex (ABC-HRP). Wash away the unbounded ABC-HRP with PBS or TBS buffer and add TMB. TMB is substrate to HRP and will be catalyzed to produce a blue color product, which changes into yellow after adding acidic stop solution. The density of the yellow product is linearly proportional to Human QPCT in the sample. Read the density of the yellow product in each well using a plate reader, and benchmark the sample wells' readings against the standard curve to determine the concentration of Human QPCT in the sample. F!!Background/Introduction: Glutaminyl-peptide cyclotransferase is an enzyme that in humans is encoded by the QPCT gene. It is mapped to 2p22.2. This gene encodes human pituitary glutaminyl cyclase, which is responsible for the presence of pyroglutamyl residues in many neuroendocrine peptides. The amino acid sequence of this enzyme is 86% identical to that of bovine glutaminyl cyclase.

NCBI Description

This gene encodes human pituitary glutaminyl cyclase, which is responsible for the presence of pyroglutamyl residues in many neuroendocrine peptides. The amino acid sequence of this enzyme is 86% identical to that of bovine glutaminyl cyclase. [provided by RefSeq, Jul 2008]

Has a bias against acidic and tryptophan residues adjacent to the N-terminal glutaminyl residue and a lack of importance of chain length after the second residue. Also catalyzes N-terminal pyroglutamate formation. In vitro, catalyzes pyroglutamate formation of N-terminally truncated form of APP amyloid-beta peptides [Glu-3]-beta-amyloid. May be involved in the N-terminal pyroglutamate formation of several amyloid-related plaque-forming peptides. Belongs to the glutaminyl-peptide cyclotransferase family. 2 isoforms of the human protein are produced by alternative splicing.
