Usage Notes
* Do not mix components from different kit lots or use reagents beyond the kit expiration date.
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Preparation and Storage
Store at 2-8 degree C.

Related Product Information for PRKCZ elisa kit
Description: This Human Protein kinase C zeta type (PRKCZ) ELISA Kit employs a two-site sandwich ELISA to quantitate PRKCZ in samples. An antibody specific for PRKCZ has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any PRKCZ present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PRKCZ is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PRKCZ bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: Protein kinase C, zeta is an enzyme which in humans is encoded by the PRKCZ gene. The PRKCZ gene encodes at least two alternative transcripts, the full-length PKC zeta and an N-terminal truncated form PKM zeta. The truncated PKMzeta is thought to be responsible for maintaining long-term memories in the brain.PKC-zeta has an N-terminal regulatory domain, followed by a hinge region and a C-terminal catalytic domain. Second messengers stimulate PKCs by binding to the regulatory domain, translocating the enzyme from cytosol to membrane, and producing a conformational change that removes autoinhibition of the PKC catalytic activity. PKM-zeta, a brain-specific isoform of PKC-zeta generated from an alternative transcript, lacks the regulatory region of full-length PKC-zeta and is therefore constitutively active.

NCBI Description

Protein kinase C (PKC) zeta is a member of the PKC family of serine/threonine kinases which are involved in a variety of cellular processes such as proliferation, differentiation and secretion. Unlike the classical PKC isoenzymes which are calcium-dependent, PKC zeta exhibits a kinase activity which is independent of calcium and diacylglycerol but not of phosphatidylserine. Furthermore, it is insensitive to typical PKC inhibitors and cannot be activated by phorbol ester. Unlike the classical PKC isoenzymes, it has only a single zinc finger module. These structural and biochemical properties indicate that the zeta subspecies is related to, but distinct from other isoenzymes of PKC. Alternative splicing results in multiple transcript variants encoding different isoforms. [provided by RefSeq, Jul 2008]

Uniprot Description

PKCZ: an AGC kinase of the PKC family. An atypical PKC: its activity is not regulated by Ca2+, PS, DAG or phorbol esters. Constitutively active.

Product Notes

The Human PRKCZ prkcz (Catalog #AAA9714810) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA9714810 ELISA Kit recognizes Human PRKCZ. It is sometimes possible for the material contained within the vial of "Protein Kinase C zeta Type (PRKCZ), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
