Usage Notes
* Do not mix components from different kit lots or use reagents beyond the kit expiration date.
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Preparation and Storage
Store at 2-8 degree C.

Related Product Information for PREP elisa kit
Description: This Human Prolyl endopeptidase (PREP) ELISA Kit employs a two-site sandwich ELISA to quantitate PREP in samples. An antibody specific for PREP has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any PREP present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PREP is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PREP bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: Prolyl endopeptidase (PE) also known as prolyl oligopeptidase or post-proline cleaving enzyme is an enzyme that in humans is encoded by the PREP gene.Prolyl endopeptidase is a large cytosolic enzyme that belongs to a distinct class of serine peptidases. It was first described in the cytosol of rabbit brain as an oligopeptidase, which degrades the nonapeptide bradykinin at the Pro-Phe bond.[5] The enzyme is involved in the maturation and degradation of peptide hormones and neuropeptides such as alpha-melanocyte-stimulating hormone, luteinizing hormone-releasing hormone (LH-RH), thyrotropin-releasing hormone, angiotensin, neurotensin, oxytocin, substance P and vasopressin. PREP cleaves peptide bonds at the C-terminal side of proline residues. Its activity is confined to action on oligopeptides of less than 10 kD and it has an absolute requirement for the trans-configuration of the peptide bond preceding proline. Prolyl endopeptidases are involved in the maturation and degradation of peptide hormones and neuropeptides.

NCBI Description

The protein encoded by this gene is a cytosolic prolyl endopeptidase that cleaves peptide bonds on the C-terminal side of prolyl residues within peptides that are up to approximately 30 amino acids long. Prolyl endopeptidases have been reported to be involved in the maturation and degradation of peptide hormones and neuropeptides. [provided by RefSeq, Jul 2008]

Uniprot Description

PREP: Cleaves peptide bonds on the C-terminal side of prolyl residues within peptides that are up to approximately 30 amino acids long. Belongs to the peptidase S9A family.

Protein type: EC 3.4.21.26; Protease

Chromosomal Location of Human Ortholog: 6q22

Cellular Component: cytoplasm; membrane

Molecular Function: protein binding; serine-type peptidase activity

Product Notes

The Human PREP prep (Catalog #AAA9714837) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA9714837 ELISA Kit recognizes Human PREP. It is sometimes possible for the material contained within the vial of "Prolyl Endopeptidase (PREP), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
