Usage Notes
* Do not mix components from different kit lots or use reagents beyond the kit expiration date.
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Preparation and Storage
Store at 2-8 degree C.

Related Product Information for PALM elisa kit
Description: This Human Paralemmin-1 (PALM) ELISA Kit employs a two-site sandwich ELISA to quantitate PALM in samples. An antibody specific for PALM has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any PALM present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PALM is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PALM bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: Paralemmin is a member of the paralemmin protein family. PALM has a highly conserved coiled-coil N terminus that displays a periodicity of acidic, basic, hydrophobic, and glutamine residues, as well as 2 potential leucine zippers. PALM has a central motif similar to a sequence found in lipid-anchored SNAREs that is involved in endoplasmic reticulum-Golgi transport. The C terminus of PALM contains a cluster of basic residues, which likely contribute to membrane association, putative palmitoylated cysteines, and a prenylation consensus motif. PALM also has phosphorylation motifs for several serine/threonine kinases. The 5-prime untranslated regions of the chicken, mouse, and human transcripts are GC rich.

NCBI Description

This gene encodes a member of the paralemmin protein family. The product of this gene is a prenylated and palmitoylated phosphoprotein that associates with the cytoplasmic face of plasma membranes and is implicated in plasma membrane dynamics in neurons and other cell types. Several alternatively spliced transcript variants have been identified, but the full-length nature of only two transcript variants has been determined. [provided by RefSeq, Jul 2008]

Uniprot Description

paralemmin: Involved in plasma membrane dynamics and cell process formation. Isoform 1 and isoform 2 are necessary for axonal and dendritic filopodia induction, for dendritic spine maturation and synapse formation in a palmitoylation-dependent manner. Belongs to the paralemmin family. 2 isoforms of the human protein are produced by alternative splicing.

Chromosomal Location of Human Ortholog: 19p13.3

Cellular Component: cytoplasmic vesicle; filopodium membrane; integral to plasma membrane; intracellular membrane-bound organelle; nucleoplasm; nucleus; plasma membrane

Molecular Function: protein binding

Biological Process: cell motility; negative regulation of adenylate cyclase activity; negative regulation of dopamine receptor signaling pathway; positive regulation of filopodium formation

Product Notes

The Human PALM palm (Catalog #AAA9714977) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA9714977 ELISA Kit recognizes Human PALM. It is sometimes possible for the material contained within the vial of "Paralemmin-1 (PALM), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
