Introduction: Human peripheral nervous system myelin contains three major proteins, designated P0, P1 and P2. The major protein, P0, is a glycoprotein, which makes up 50% of the total peripheral nervous system myelin protein. It has a molecular weight of 30,000 and is distinct from two basic proteins, P1 and P2, with molecular weights of 18,000 and 13,000, respectively. The P0 protein is unique to peripheral nervous system myelin and is not present in central nervous system myelin. The P0 protein presumably plays a major structural role in the myelin sheath. P0 protein is composed of an intracellular, a transmembrane, and an extracellular domain that is an immunoglobulin-like domain. Antibodies against the myelin P0 protein have been detected in patients with Guilllain-Barre syndrome and in patients with peripheral neuropathy. In animal models, experimental autoimmune neuritis has been induced by immunization against heterologous P0 protein, suggesting that myelin P0 protein may be a potential autoantigen involved in etiology of autoimmune diseases.

Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with purified myelin P0 antigen. Samples are then added to the appropriate microtiter plate wells and incubated. Then add Horseradish Peroxidase (HRP)-conjugated anti-human IgG to each well and incubate. Finally, a TMB (3,3'5, 5' tetramethyl-benzidine) substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. Calculate the valence of myelin P0 antibody(IgG) in the samples.
