• Test Principle: This assay employs the competitive inhibition enzyme immunoassay technique. A monoclonal antibody specific to Glutathione (GSH) has been pre-coated onto a microplate. A competitive inhibition reaction is launched between biotin labeled Glutathione (GSH) and unlabeled Glutathione (GSH)(Standards or samples) with the pre-coated antibody specific to Glutathione (GSH). After incubation the unbound conjugate is washed off. Next, avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. The amount of bound HRP conjugate is reverse proportional to the concentration of Glutathione (GSH) in the sample. After addition of the substrate solution, the intensity of color developed is reverse proportional to the concentration of Glutathione (GSH) in the sample.
• Assay Procedure Summary: 1. Prepare all reagents, samples and standards; 2. Add 100ul standard or sample to each well. Incubate 2 hours at 37 degree C; 3. Aspirate and add 100ul prepared Detection Reagent A. Incubate 1 hour at 37 degree C; 4. Aspirate and wash 3 times; 5. Add 100ul prepared Detection Reagent B. Incubate 30 minutes at 37 degree C; 6. Aspirate and wash 5 times; 7. Add 90ul Substrate Solution. Incubate 10-20 minutes at 37 degree C; 8. Add 50ul Stop Solution. Read at 450nm immediately.
• Preparation and Storage: Store at 4 degree C for 1 month; -20 degree C for long-term (within shelf life); The stability of kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. To minimize extra influence on the performance, operation procedures and lab conditions, especially room temperature, air humidity, incubator temperature should be strictly controlled. It is also strongly suggested that the whole assay is performed by the same operator from the beginning to the end.

NCBI Description

This gene encodes a protein which contains a C-terminal domain able to interact with the angiotension II (AT2) receptor and a large coiled-coil region allowing dimerization. Multiple alternatively spliced transcript variants encoding different isoforms have been found for this gene. One of the transcript variants has been shown to encode a mitochondrial protein that acts as a tumor suppressor and partcipates in AT2 signaling pathways. Other variants may encode nuclear or transmembrane proteins but it has not been determined whether they also participate in AT2 signaling pathways. [provided by RefSeq, Jul 2008]

Cooperates with AGTR2 to inhibit ERK2 activation and cell proliferation. May be required for AGTR2 cell surface expression. Together with PTPN6, induces UBE2V2 expression upon angiotensin-II stimulation. Isoform 1 inhibits breast cancer cell proliferation, delays the progression of mitosis by prolonging metaphase and reduces tumor growth.
