Usage Notes
* Do not mix components from different kit lots or use reagents beyond the kit expiration date.
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Preparation and Storage
Store at 2-8 degree C.

Related Product Information for FIS1 elisa kit
Description: This Human Mitochondrial fission 1 protein (FIS1) ELISA Kit employs a two-site sandwich ELISA to quantitate FIS1 in samples. An antibody specific for FIS1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any FIS1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for FIS1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of FIS1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: The balance between fission and fusion regulates the morphology of mitochondria. TTC11 is a component of a mitochondrial complex that promotes mitochondrial fission. The deduced 152-amino acid protein contains a central leucine zipper, a coiled-coil region, and a C-terminal transmembrane domain. It also contains a region showing high interspecies homology and a putative tetratricopeptide repeat. Immunocytochemical analysis of HeLa cells detected endogenous FIS1 expression in mitochondria. Protease treatment indicated that FIS1 is expressed at the outer mitochondrial membrane. Transfection of a C-terminal truncated FIS1 resulted in cytoplasmic expression, indicating that the C terminus is required for mitochondrial localization.

NCBI Description

The balance between fission and fusion regulates the morphology of mitochondria. TTC11 is a component of a mitochondrial complex that promotes mitochondrial fission (James et al., 2003 [PubMed 12783892]).[supplied by OMIM, Mar 2008]

Uniprot Description

FIS1: Promotes the fragmentation of the mitochondrial network and its perinuclear clustering. Can induce cytochrome c release from the mitochondrion to the cytosol, ultimately leading to apoptosis. Also mediates peroxisomal fission. Belongs to the FIS1 family.

Protein type: Membrane protein, integral; Mitochondrial

Chromosomal Location of Human Ortholog: 7q22.1

Cellular Component: integral to peroxisomal membrane; protein complex; mitochondrion; membrane; peroxisome; integral to mitochondrial outer membrane

Molecular Function: protein binding; receptor binding

Biological Process: peroxisome fission; mitochondrial fission; release of cytochrome c from mitochondria; elevation of cytosolic calcium ion concentration; mitochondrial fusion; reduction of endoplasmic reticulum calcium ion concentration; mitochondrion degradation; positive regulation of caspase activity; mitochondrial fragmentation during apoptosis; protein targeting to mitochondrion; protein homooligomerization; elevation of mitochondrial calcium ion concentration

Product Notes

The Human FIS1 fis1 (Catalog #AAA9715918) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA9715918 ELISA Kit recognizes Human FIS1. It is sometimes possible for the material contained within the vial of "Mitochondrial Fission 1 Protein (FIS1), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
