Intended Uses: The LF test kit is a highly sensitive two-site enzyme linked immunoassay (ELISA) for measuring LF in human biological samples. If the ELISA is to be used outside the intended use, the user may need to optimize for said use.

Principle of the Assay: The principle of the double antibody sandwich ELISA is represented in Figure 1. In this assay the lactoferrin (LF) present in samples reacts with the anti-LF antibodies, which have been adsorbed to the surface of polystyrene microtitre wells. After the removal of unbound proteins by washing, anti-LF antibodies conjugated with horseradish peroxidase (HRP) are added. These enzyme-labeled antibodies form complexes with the previously bound LF. Following another washing step, the enzyme bound to the immunosorbent is assayed by the addition of a chromogenic substrate, 3,3',5,5'-tetramethylbenzidine (TMB). The quantity of bound enzyme varies directly with the concentration of LF in the sample tested; thus, the absorbance, at 450 nm, is a measure of the concentration of LF in the test sample. The quantity of LF in the test sample can be interpolated from the standard curve constructed from the standards, and corrected for sample dilution.

NCBI Description

This gene is a member of the transferrin family of genes and its protein product is found in the secondary granules of neutrophils. The protein is a major iron-binding protein in milk and body secretions with an antimicrobial activity, making it an important component of the non-specific immune system. The protein demonstrates a broad spectrum of properties, including regulation of iron homeostasis, host defense against a broad range of microbial infections, anti-inflammatory activity, regulation of cellular growth and differentiation and protection against cancer development and metastasis. Alternatively spliced transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq]
