Usage Notes
* Do not mix components from different kit lots or use reagents beyond the kit expiration date.
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Preparation and Storage
Store at 2-8 degree C.

Related Product Information for AADAT elisa kit
Description: This Human Kynurenine/alpha-aminoadipate aminotransferase, mitochondrial (AADAT) ELISA Kit employs a two-site sandwich ELISA to quantitate AADAT in samples. An antibody specific for AADAT has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any AADAT present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for AADAT is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of AADAT bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: Aminoadipate Aminotransferase is highly similar to mouse and rat kynurenine aminotransferase II. The rat protein is a homodimer with two transaminase activities. One activity is the transamination of alpha-aminoadipic acid, a final step in the saccaropine pathway which is the major pathway for L-lysine catabolism. The other activity involves the transamination of kynurenine to produce kynurenine acid, the precursor of kynurenic acid which has neuroprotective properties. Two alternative transcripts encoding the same isoform have been identified, however, additional alternative transcripts and isoforms may exist. Higher expression in the liver. Also found in heart, brain, kidney, pancreas, prostate, testis and ovary.

NCBI Description

This gene encodes a protein that is highly similar to mouse and rat kynurenine aminotransferase II. Several transcript variants encoding two different isoforms have been found for this gene. [provided by RefSeq, Nov 2013]

Uniprot Description

AADAT: Transaminase with broad substrate specificity. Has transaminase activity towards aminoadipate, kynurenine, methionine and glutamate. Shows activity also towards tryptophan, aspartate and hydroxykynurenine. Accepts a variety of oxo-acids as amino- group acceptors, with a preference for 2-oxoglutarate, 2- oxocaproic acid, phenylpyruvate and alpha-oxo-gamma-methiol butyric acid. Can also use glyoxylate as amino-group acceptor (in vitro). Belongs to the class-I pyridoxal-phosphate-dependent aminotransferase family. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: EC 2.6.1.7; Amino Acid Metabolism - tryptophan; EC 2.6.1.39; Amino Acid Metabolism - lysine degradation; Transferase; Amino Acid Metabolism - lysine biosynthesis; Mitochondrial

Chromosomal Location of Human Ortholog: 4q33

Cellular Component: mitochondrial matrix

Molecular Function: protein homodimerization activity; kynurenine-oxoglutarate transaminase activity; 2-aminoadipate transaminase activity; pyridoxal phosphate binding

Biological Process: glutamate metabolic process; tryptophan catabolic process to kynurenine; L-lysine catabolic process to acetyl-CoA via saccharopine; lysine catabolic process; biosynthetic process; tryptophan catabolic process; 2-oxoglutarate metabolic process

Product Notes

The Human AADAT aadat (Catalog #AAA9714509) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA9714509 ELISA Kit recognizes Human AADAT. It is sometimes possible for the material contained within the vial of "Kynurenine/alpha-Aminoadipate Aminotransferase, Mitochondrial (AADAT), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
