Related Product Information for FURIN elisa kit
Principle of the assay: human Furin ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for Furin has been precoated onto 96-well plates. Standards(NSO, D108-E715) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for Furin is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human Furin amount of sample captured in plate.
Background: Furin(Fur), also known as PACE, is a protein that in humans is encoded by the FURIN gene. The protein encoded by this gene is an enzyme which belongs to the subtilisin-like proprotein convertase family. The FUR gene is located approximately 1 kb upstream of the FES gene on chromosome 15q25-q26. This gene is thought to play a role in tumor progression. Furin is one of the proteases responsible for the proteolytic cleavage of HIV envelope polyprotein precursor gp160 to gp120 and gp41 prior to viral assembly. Furin is enriched in the Golgi apparatus, where it functions to cleave other proteins into their mature/active forms. Expression of furin in T-cells is required for maintenance of peripheral immune tolerance

NCBI Description

This gene encodes a member of the subtilisin-like proprotein convertase family, which includes proteases that process protein and peptide precursors trafficking through regulated or constitutive branches of the secretory pathway. It encodes a type 1 membrane bound protease that is expressed in many tissues, including neuroendocrine, liver, gut, and brain. The encoded protein undergoes an initial autocatalytic processing event in the ER and then sorts to the trans-Golgi network through endosomes where a second autocatalytic event takes place and the catalytic activity is acquired. The product of this gene is one of the seven basic amino acid-specific members which cleave their substrates at single or paired basic residues. Some of its substrates include proparathyroid hormone, transforming growth factor beta 1 precursor, proalbumin, pro-beta-secretase, membrane type-1 matrix metalloproteinase, beta subunit of pro-nerve growth factor and von Willebrand factor. It is also thought to be one of the proteases responsible for the activation of HIV envelope glycoproteins gp160 and gp140 and may play a role in tumor progression. This gene is located in close proximity to family member proprotein convertase subtilisin/kexin type 6 and upstream of the FES oncogene. Alternative splicing results in multiple transcript variants. Product Notes

The Human FURIN furin (Catalog #AAA176510) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA176510 ELISA Kit recognizes Human FURIN. It is sometimes possible for the material contained within the vial of "Furin, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
