The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Eukaryotic Translation Initiation Factor 5A2 (EIF5A2). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Eukaryotic Translation Initiation Factor 5A2 (EIF5A2). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Eukaryotic Translation Initiation Factor 5A2 (EIF5A2), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Eukaryotic Translation Initiation Factor 5A2 (EIF5A2) in the samples is then determined by comparing the O.D. of the samples to the standard curve.

EIF5A2: mRNA-binding protein involved in translation elongation. Has an important function at the level of mRNA turnover, probably acting downstream of decapping. Involved in actin dynamics and cell cycle progression, mRNA decay and probably in a pathway involved in stress response and maintenance of cell wall integrity. Functions as a regulator of apoptosis. Mediates effects of polyamines on neuronal process extension and survival. May play an important role in brain development and function, and in skeletal muscle stem cell differentiation. Belongs to the eIF-5A family.
