Intended Uses: This sandwich kit is for the accurate quantitative detection of Human Thyroxine 5-deiodinase (also known as DIO3) in serum, plasma, cell culture supernates, Ascites, tissue homogenates or other biological fluids.
Principle of the Assay: This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with Human DIO3 antibody. DIO3 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Human DIO3 Antibody is added and binds to DIO3 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated DIO3 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Human DIO3. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

The protein encoded by this intronless gene belongs to the iodothyronine deiodinase family. It catalyzes the inactivation of thyroid hormone by inner ring deiodination of the prohormone thyroxine (T4) and the bioactive hormone 3,3',5-triiodothyronine (T3) to inactive metabolites, 3,3',5'-triiodothyronine (RT3) and 3,3'-diiodothyronine (T2), respectively. As in other verterbrates, studies in xenopus show that this enzyme is involved in early embryonic development. This protein is a selenoprotein, containing the rare selenocysteine (Sec) amino acid at its active site. Sec is encoded by the UGA codon, which normally signals translation termination. The 3' UTRs of selenoprotein mRNAs contain a conserved stem-loop structure, designated the Sec insertion sequence (SECIS) element, that is necessary for the recognition of UGA as a Sec codon rather than as a stop signal.
