Human Cytochrome b 245 light chain (CYBA) ELISA Kit
Principle of the Assay: CYBA ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-CYBA antibody and an CYBA-HRP conjugate. The assay sample and buffer are incubated together with CYBA-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CYBA concentration since CYBA from samples and CYBA-HRP conjugate compete for the anti-CYBA antibody binding site. Since the number of sites is limited, as more sites are occupied by CYBA from the sample, fewer sites are left to bind CYBA-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CYBA concentration in each sample is interpolated from this standard curve.

Cytochrome b is comprised of a light chain (alpha) and a heavy chain (beta). This gene encodes the light, alpha subunit which has been proposed as a primary component of the microbicidal oxidase system of phagocytes. Mutations in this gene are associated with autosomal recessive chronic granulomatous disease (CGD), that is characterized by the failure of activated phagocytes to generate superoxide, which is important for the microbicidal activity of these cells. Defects in CYBA are a cause of chronic granulomatous disease autosomal recessive cytochrome-b-negative (ARCGD). Chronic granulomatous disease is a genetically heterogeneous disorder characterized by the inability of neutrophils and phagocytes to kill microbes that they have ingested. Patients suffer from life-threatening bacterial/fungal infections. Belongs to the p22phox family.
