Principle of the assay: CSTD ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-CSTD antibody and an CSTD-HRP conjugate. The assay sample and buffer are incubated together with CSTD-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CSTD concentration since CSTD from samples and CSTD-HRP conjugate compete for the anti-CSTD antibody binding site. Since the number of sites is limited, as more sites are occupied by CSTD from the sample, fewer sites are left to bind CSTD-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CSTD concentration in each sample is interpolated from this standard curve.

NCBI Description

The cystatin superfamily encompasses proteins that contain multiple cystatin-like sequences. Some of the members are active cysteine protease inhibitors, while others have lost or perhaps never acquired this inhibitory activity. There are three inhibitory families in the superfamily, including the type 1 cystatins (stefins), type 2 cystatins and the kininogens. The type 2 cystatin proteins are a class of cysteine proteinase inhibitors found in a variety of human fluids and secretions. The cystatin locus on chromosome 20 contains the majority of the type 2 cystatin genes and pseudogenes. This gene is located in the cystatin locus and encodes a protein found in saliva and tears. The encoded protein may play a protective role against proteinases present in the oral cavity. [provided by RefSeq, Jul 2008]

Uniprot Description

CST5: Cysteine proteinase inhibitor that possibly plays a protective role against proteinases present in the oral cavity. The order of preference for inhibition is cathepsin S > cathepsin H > cathepsin L > cathepsin B. Belongs to the cystatin family.

Protein type: Secreted, signal peptide; Secreted

Chromosomal Location of Human Ortholog: 20p11.21

Product Notes

The Human CST5 cst5 (Catalog #AAA7225016) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA7225016 ELISA Kit recognizes Human CST5. It is sometimes possible for the material contained within the vial of "Cystatin D (CST5), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
