Principle of the Assay: The Picokine Human CST1 Pre-Coated ELISA (Enzyme-Linked Immunosorbent Assay) kit is a solid phase immunoassay specially designed to measure Human CST1 with a 96-well strip plate that is pre-coated with antibody specific for CST1. The detection antibody is a biotinylated antibody specific for CST1. The capture antibody is a monoclonal antibody from mouse, the detection antibody is a biotinylated polyclonal antibody from goat. The kit contains recombinant Human CST1 with immunogen: Expression system for standard: NSO, Immunogen sequence: W21-S141. The kit is analytically validated with ready to use reagents. To measure Human CST1, add standards and samples to the wells, then add the biotinylated detection antibody. Wash the wells with PBS or TBS buffer, and add Avidin-Biotin-Peroxidase Complex (ABC-HRP). Wash away the unbounded ABC-HRP with PBS or TBS buffer and add TMB. TMB is substrate to HRP and will be catalyzed to produce a blue color product, which changes into yellow after adding acidic stop solution. The density of the yellow product is linearly propotional to Human CST1 in the sample. Read the density of the yellow product in each well using a plate reader, and benchmark the sample wells' readings against the standard curve to determine the concentration of Human CST1 in the sample!!Background/Introduction: Cystatin-SN is a protein that in humans is encoded by the CST1 gene. The cystatin superfamily encompasses proteins that contain multiple cystatin-like sequences. Some of the members are active cysteine protease inhibitors, while others have lost or perhaps never acquired this inhibitory activity. There are three inhibitory families in the superfamily, including the type 1 cystatins (stefins), type 2 cystatins and the kininogens. The type 2 cystatin proteins are a class of cysteine proteinase inhibitors found in a variety of human fluids and secretions, where they appear to provide protective functions. The cystatin locus on chromosome 20 contains the majority of the type 2 cystatin genes and pseudogenes. This gene is located in the cystatin locus and encodes a cysteine proteinase inhibitor found in saliva, tears, urine, and seminal fluid.

NCBI Description

The encoded protein may play a protective role against proteinases present in the oral cavity. [provided by RefSeq, Jul 2008]

Uniprot Description

Cysteine proteinase inhibitor that possibly plays a protective role against proteinases present in the oral cavity. The order of preference for inhibition is cathepsin S > cathepsin H > cathepsin L > cathepsin B.

Product Notes

The Human There is no detectable cross-reactivity with other relevant proteins. CST5 cst5 (Catalog #AAA1751708) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA1751708 ELISA Kit recognizes Human There is no detectable cross-reactivity with other relevant proteins. CST5. It is sometimes possible for the material contained within the vial of "Cystatin D/CST5, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
