Intended Uses: This sandwich kit is for the accurate quantitative detection of Human Cartilage intermediate layer protein 1 (also known as CILP) in serum, plasma, cell culture supernates, Ascites, tissue homogenates or other biological fluids.

Principle of the Assay: This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with Human CILP antibody. CILP present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Human CILP Antibody is added and binds to CILP in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated CILP antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Human CILP. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Uniprot Description

CILP: Probably plays a role in cartilage scaffolding. May act by antagonizing TGF-beta1 (TGFB1) and IGF1 functions. Has the ability to suppress IGF1-induced proliferation and sulfated proteoglycan synthesis, and inhibits ligand-induced IGF1R autophosphorylation. May inhibit TGFB1-mediated induction of cartilage matrix genes via its interaction with TGFB1. Overexpression may lead to impair chondrocyte growth and matrix repair and indirectly promote inorganic pyrophosphate (PPi) supersaturation in aging and osteoarthritis cartilage. Defects in CILP are a cause of susceptibility to intervertebral disk disease (IDD). A common musculo- skeletal disorder caused by degeneration of intervertebral disks of the lumbar spine. It results in low-back pain and unilateral leg pain. Susceptibility to intervertebral disk disease, is conferred by variant p.Ile395Thr (PubMed:15864306).
