Human Cannabinoid receptor 2 (CNR2) ELISA Kit
Principle of the Assay: CNR2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-CNR2 antibody and an CNR2-HRP conjugate. The assay sample and buffer are incubated together with CNR2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CNR2 concentration since CNR2 from samples and CNR2-HRP conjugate compete for the anti-CNR2 antibody binding site. Since the number of sites is limited, as more sites are occupied by CNR2 from the sample, fewer sites are left to bind CNR2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CNR2 concentration in each sample is interpolated from this standard curve.

The cannabinoid delta-9-tetrahydrocannabinol is the principal psychoactive ingredient of marijuana. The proteins encoded by this gene and the cannabinoid receptor 1 (brain) (CNR1) gene have the characteristics of a guanine nucleotide-binding protein (G-protein)-coupled receptor for cannabinoids. They inhibit adenylate cyclase activity in a dose-dependent, stereoselective, and pertussis toxin-sensitive manner. These proteins have been found to be involved in the cannabinoid-induced CNS effects (including alterations in mood and cognition) experienced by users of marijuana.
