Intended Uses: This CDH16 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human CDH16. This ELISA kit for research use only!

Principle of the Assay||CDH16 ELISA kit applies the competitive enzyme immunoassay technique utilizing a Polyclonal anti-CDH16 antibody and an CDH16-HRP conjugate. The assay sample and buffer are incubated together with CDH16-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CDH16 concentration since CDH16 from samples and CDH16-HRP conjugate compete for the anti-CDH16 antibody binding site. Since the number of sites is limited, as more sites are occupied by CDH16 from the sample, fewer sites are left to bind CDH16-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CDH16 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene is a member of the cadherin superfamily, genes encoding calcium-dependent, membrane-associated glycoproteins. Mapped to a previously identified cluster of cadherin genes on chromosome 16q22.1, the gene localizes with superfamily members CDH1, CDH3, CDH5, CDH8 and CDH11. The protein consists of an extracellular domain containing 6 cadherin domains, a transmembrane region and a truncated cytoplasmic domain but lacks the prosequence and tripeptide HAV adhesion recognition sequence typical of most classical cadherins. Expression is exclusively in kidney, where the protein functions as the principal mediator of homotypic cellular recognition, playing a role in the morphogenic direction of tissue development. Alternatively spliced transcript variants encoding distinct isoforms have been identified. [provided by RefSeq, Mar 2011]
