Usage Notes
* Do not mix components from different kit lots or use reagents beyond the kit expiration date.
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Preparation and Storage
Store at 2-8 degree C.

Description: Human beta-hCG ELISA Kit employs a two-site sandwich ELISA to quantitate beta-hCG in samples. An antibody specific for beta-hCG has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any beta-hCG present is bound by the immobilized antibody. After removing any unbound substances, a HRP conjugated antibody specific for beta-hCG is added to the wells. Following a wash to remove any unbound enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of beta-hCG bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: Human chorionic gonadotropin (hCG) is a glycoprotein hormone produced by trophoblastic cells of the placenta beginning 10 to 12 days after conception. Maintenance of the fetus in the first trimester of pregnancy requires the production of hCG, which binds to the corpus luteum of the ovary which is stimulated to produce progesterone which in turn maintains the secretory endometrium. hCG is present only in trace amounts in non pregnant urine and sera. It rises sharply during pregnancy. HCG is composed of two non identical, non covalently linked polypeptide chains designated as the a and b subunits. The a subunit of HCG is nearly identical to that of thyroid stimulating hormone (TSH), follicle stimulating hormone (FSH) and luteinizing hormone (LH). A germ cell tumor which is positive for cytokeratin, placental alkaline phosphatase (PLAP) and HCG but negative for EMA and AFP is probably a choriocarcinoma.

NCBI Description

This gene is a member of the glycoprotein hormone beta chain family and encodes the beta 3 subunit of chorionic gonadotropin (CG). Glycoprotein hormones are heterodimers consisting of a common alpha subunit and an unique beta subunit which confers biological specificity. CG is produced by the trophoblastic cells of the placenta and stimulates the ovaries to synthesize the steroids that are essential for the maintenance of pregnancy. The beta subunit of CG is encoded by 6 genes which are arranged in tandem and inverted pairs on chromosome 19q13.3 and contiguous with the luteinizing hormone beta subunit gene. Beta subunit of the human chorionic gonadotropin (hCG). hCG is a complex glycoprotein composed of two glycosylated subunits alpha and beta which are non-covalently associated. The alpha subunit is identical to those in the pituitary gonadotropin hormones (LH, FSH and TSH). The beta subunits are distinct in each of the hormones and confer receptor and biological specificity. Has an essential role in pregnancy and maternal adaptation. Stimulates the ovaries to synthesize the steroids that are essential for the maintenance of pregnancy.
