Intended Uses: This ADRBK1 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human ADRBK1. This ELISA kit for research use only!

Principle of the Assay: ADRBK1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-ADRBK1 antibody and an ADRBK1-HRP conjugate. The assay sample and buffer are incubated together with ADRBK1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ADRBK1 concentration since ADRBK1 from samples and ADRBK1-HRP conjugate compete for the anti-ADRBK1 antibody binding site. Since the number of sites is limited, as more sites are occupied by ADRBK1 from the sample, fewer sites are left to bind ADRBK1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ADRBK1 concentration in each sample is interpolated from this standard curve.

NCBI Description

The product of this gene phosphorylates the beta-2-adrenergic receptor and appears to mediate agonist-specific desensitization observed at high agonist concentrations. This protein is an ubiquitous cytosolic enzyme that specifically phosphorylates the activated form of the beta-adrenergic and related G-protein-coupled receptors. Abnormal coupling of beta-adrenergic receptor to G protein is involved in the pathogenesis of the failing heart. [provided by RefSeq, Jul 2008]

Appears to mediate agonist-specific desensitization observed at high agonist concentrations. Expression level is consistently elevated in chronic human heart failure. Mouse models of severe heart failure have been used to demonstrate that inhibition of BARK1 with a peptide inhibitor is sufficient to increase mean survival, reduce dialation and improve cardiac function.
