Reactivity
Human
Specificity
This assay has high sensitivity and excellent specificity for detection of AFM. No significant cross-reactivity or interference between AFM and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between AFM and all the analogues, therefore, cross reaction may still exist in some cases.
Samples
Serum, plasma, cell culture supernatants, body fluid and tissue homogenate
Assay Type
Quantitative Competitive
Sensitivity
1.0 ug/mL
Preparation and Storage
Store all reagents at 2-8 degree C.

Intended Uses: This AFM ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human AFM. This ELISA kit for research use only!

Principle of the Assay: AFM ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-AFM antibody and an AFM-HRP conjugate. The assay sample and buffer are incubated together with AFM-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the AFM concentration since AFM from samples and AFM-HRP conjugate compete for the anti-AFM antibody binding site. Since the number of sites is limited, as more sites are occupied by AFM from the sample, fewer sites are left to bind AFM-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The AFM concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene is a member of the albumin gene family, which is comprised of four genes that localize to chromosome 4 in a tandem arrangement. These four genes encode structurally-related serum transport proteins that are known to be evolutionarily related. The protein encoded by this gene is regulated developmentally, expressed in the liver and secreted into the bloodstream. [provided by RefSeq, Jul 2008]

Uniprot Description

A-albumin: Vitamin E binding protein. May transport vitamin E in body fluids under conditions where the lipoprotein system is not sufficient. May be involved in the regulation and transport of vitamin E at the blood-brain barrier. Belongs to the ALB/AFP/VDB family.

Protein type: Secreted, signal peptide; Secreted; Carrier

Chromosomal Location of Human Ortholog: 4q13.3

Cellular Component: extracellular space; extracellular region

Molecular Function: vitamin E binding

Biological Process: vitamin transport

Product Notes

The Human AFM afm (Catalog #AAA7217736) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA7217736 ELISA Kit recognizes Human AFM. It is sometimes possible for the material contained within the vial of "Afamin (AFM), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
