Human Acrosin binding protein (ACRBP) ELISA Kit
Principle of the Assay: ACRBP ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-ACRBP antibody and an ACRBP-HRP conjugate. The assay sample and buffer are incubated together with ACRBP-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ACRBP concentration since ACRBP from samples and ACRBP-HRP conjugate compete for the anti-ACRBP antibody binding site. Since the number of sites is limited, as more sites are occupied by ACRBP from the sample, fewer sites are left to bind ACRBP-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ACRBP concentration in each sample is interpolated from this standard curve.

The protein encoded by this gene is similar to proacrosin binding protein sp32 precursor found in mouse, guinea pig, and pig. This protein is located in the sperm acrosome and is thought to function as a binding protein to proacrosin for packaging and condensation of the acrosin zymogen in the acrosomal matrix. This protein is a member of the cancer/testis family of antigens and it is found to be immunogenic. In normal tissues, this mRNA is expressed only in testis, whereas it is detected in a range of different tumor types such as bladder, breast, lung, liver, and colon. [provided by RefSeq, Jul 2008]
