Intended Uses: This RPL14 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human RPL14. This ELISA kit for research use only!

Principle of the Assay: RPL14 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-RPL14 antibody and an RPL14-HRP conjugate. The assay sample and buffer are incubated together with RPL14-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the RPL14 concentration since RPL14 from samples and RPL14-HRP conjugate compete for the anti-RPL14 antibody binding site. Since the number of sites is limited, as more sites are occupied by RPL14 from the sample, fewer sites are left to bind RPL14-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The RPL14 concentration in each sample is interpolated from this standard curve.

NCBI Description

Ribosomes, the organelles that catalyze protein synthesis, consist of a small 40S subunit and a large 60S subunit. Together these subunits are composed of 4 RNA species and approximately 80 structurally distinct proteins. This gene encodes a ribosomal protein that is a component of the 60S subunit. The protein belongs to the L14E family of ribosomal proteins. It contains a basic region-leucine zipper (bZIP)-like domain. The protein is located in the cytoplasm. This gene contains a trinucleotide (GCT) repeat tract whose length is highly polymorphic; these triplet repeats result in a stretch of alanine residues in the encoded protein. Transcript variants utilizing alternative polyA signals and alternative 5'-terminal exons exist but all encode the same protein. As is typical for genes encoding ribosomal proteins, there are multiple processed pseudogenes of this gene dispersed through the genome. [provided by RefSeq, Jul 2008]
