Human 60S acidic ribosomal protein P2 (RPLP2) ELISA Kit
Principle of the Assay: RPLP2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-RPLP2 antibody and an RPLP2-HRP conjugate. The assay sample and buffer are incubated together with RPLP2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the RPLP2 concentration since RPLP2 from samples and RPLP2-HRP conjugate compete for the anti-RPLP2 antibody binding site. Since the number of sites is limited, as more sites are occupied by RPLP2 from the sample, fewer sites are left to bind RPLP2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The RPLP2 concentration in each sample is interpolated from this standard curve.

Ribosomes, the organelles that catalyze protein synthesis, consist of a small 40S subunit and a large 60S subunit. Together these subunits are composed of 4 RNA species and approximately 80 structurally distinct proteins. This gene encodes a ribosomal phosphoprotein that is a component of the 60S subunit. The protein, which is a functional equivalent of the E. coli L7/L12 ribosomal protein, belongs to the L12P family of ribosomal proteins. It plays an important role in the elongation step of protein synthesis. Unlike most ribosomal proteins, which are basic, the encoded protein is acidic. Its C-terminal end is nearly identical to the C-terminal ends of the ribosomal phosphoproteins P0 and P1. The P2 protein can interact with P0 and P1 to form a pentameric complex consisting of P1 and P2 dimers, and a P0 monomer. The protein is located in the cytoplasm. As is typical for genes encoding ribosomal proteins, there are multiple processed pseudogenes of this gene dispersed through the genome. [provided by RefSeq, Jul 2008]
