Human 40S ribosomal protein S3 (RPS3) ELISA Kit
Principle of the Assay: RPS3 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-RPS3 antibody and an RPS3-HRP conjugate. The assay sample and buffer are incubated together with RPS3-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the RPS3 concentration since RPS3 from samples and RPS3-HRP conjugate compete for the anti-RPS3 antibody binding site. Since the number of sites is limited, as more sites are occupied by RPS3 from the sample, fewer sites are left to bind RPS3-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The RPS3 concentration in each sample is interpolated from this standard curve.

Ribosomes, the organelles that catalyze protein synthesis, consist of a small 40S subunit and a large 60S subunit. Together these subunits are composed of 4 RNA species and approximately 80 structurally distinct proteins. This gene encodes a ribosomal protein that is a component of the 40S subunit, where it forms part of the domain where translation is initiated. The protein belongs to the S3P family of ribosomal proteins. Studies of the mouse and rat proteins have demonstrated that the protein has an extraribosomal role as an endonuclease involved in the repair of UV-induced DNA damage. The protein appears to be located in both the cytoplasm and nucleus but not in the nucleolus. Higher levels of expression of this gene in colon adenocarcinomas and adenomatous polyps compared to adjacent normal colonic mucosa have been observed. This gene is co-transcribed with the small nucleolar RNA genes U15A and U15B, which are located in its first and fifth introns, respectively. As is typical for genes encoding ribosomal proteins, there are multiple processed pseudogenes of this gene dispersed through the genome.
