Test Principle
The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Poly ADP Ribose Polymerase (PARP). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Poly ADP Ribose Polymerase (PARP). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Poly ADP Ribose Polymerase (PARP), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of Poly ADP Ribose Polymerase (PARP) in the samples is then determined by comparing the O.D. of the samples to the standard curve.
Preparation and Storage
Store at 4 degree C for 1 month/ -20 degree C for long-term (within shelf life).
The stability of kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. To minimize extra influence on the performance, operation procedures and lab conditions, especially room temperature, air humidity, incubator temperature should be strictly controlled. It is also strongly suggested that the whole assay is performed by the same operator from the beginning to the end.

Uniprot Description

IGHD: IgD is the major antigen receptor isotype on the surface of most peripheral B-cells, where it is coexpressed with IgM. The membrane-bound IgD (mIgD) induces the phosphorylation of CD79A and CD79B by the Src family of protein tyrosine kinases. Soluble IgD (sIgD) concentration in serum below those of IgG, IgA, and IgM but much higher than that of IgE. IgM and IgD molecules present on B cells have identical V regions and antigen-binding sites. After the antigen binds to the B-cell receptor, the secreted form sIgD is shut off. IgD is a potent inducer of TNF, IL1B, and IL1RN. IgD also induces release of IL6, IL10, and LIF from peripheral blood mononuclear cells. Monocytes seem to be the main producers of cytokines in vitro in the presence of IgD. 2 isoforms of the human protein are produced by alternative splicing.

Product Notes

The Human IgD ighd (Catalog #AAA2709665) is a CLIA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA2709665 CLIA Kit recognizes Human IgD. AAA Biotech's Immunoglobulin D (IgD) can be used in a range of immunoassay formats including, but not limited to, Chemiluminescence Immunoassay (CLIA). Chemiluminescent immunoassay for Antigen Detection. Researchers should empirically determine the suitability of the IgD ighd for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Immunoglobulin D (IgD), CLIA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
