Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate HSP-40 in samples. An antibody specific for HSP-40 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyHSP-40 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for HSP-40 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of HSP-40 bound in the initial step. The color development is stopped and the intensity of the color is measured.

NCBI Description

This gene encodes a member of the DnaJ or Hsp40 (heat shock protein 40 kD) family of proteins. DNAJ family members are characterized by a highly conserved amino acid stretch called the 'J-domain' and function as one of the two major classes of molecular chaperones involved in a wide range of cellular events, such as protein folding and oligomeric protein complex assembly. The encoded protein is a molecular chaperone that stimulates the ATPase activity of Hsp70 heat-shock proteins in order to promote protein folding and prevent misfolded protein aggregation.
