Intended Uses: This HNF-4-alpha ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human HNF-4-alpha. This ELISA kit for research use only!

Principle of the Assay: HNF-4-alpha ELISA kit applies the competitive enzyme immunoassay technique utilizing a Polyclonal anti-HNF-4-alpha antibody and an HNF-4-alpha-HRP conjugate. The assay sample and buffer are incubated together with HNF-4-alpha-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the HNF-4-alpha concentration since HNF-4-alpha from samples and HNF-4-alpha-HRP conjugate compete for the anti-HNF-4-alpha antibody binding site. Since the number of sites is limited, as more sites are occupied by HNF-4-alpha from the sample, fewer sites are left to bind HNF-4-alpha-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The HNF-4-alpha concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene is a nuclear transcription factor which binds DNA as a homodimer. The encoded protein controls the expression of several genes, including hepatocyte nuclear factor 1 alpha, a transcription factor which regulates the expression of several hepatic genes. This gene may play a role in development of the liver, kidney, and intestines. Mutations in this gene have been associated with monogenic autosomal dominant non-insulin-dependent diabetes mellitus type I. Alternative splicing of this gene results in multiple transcript variants encoding several different isoforms. [provided by RefSeq, Apr 2012]
