Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate HIST2H2BD in samples. An antibody specific for HIST2H2BD has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyHIST2H2BD present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for HIST2H2BD is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of HIST2H2BD bound in the initial step. The color development is stopped and the intensity of the color is measured.

NCBI Description

Histones are basic nuclear proteins responsible for nucleosome structure of the chromosomal fiber in eukaryotes. Two molecules of each of the four core histones (H2A, H2B, H3, and H4) form an octamer, around which approximately 146 bp of DNA is wrapped in repeating units, called nucleosomes. The linker histone, H1, interacts with linker DNA between nucleosomes and functions in the compaction of chromatin into higher order structures. This gene is found in a histone cluster on chromosome 1. [provided by RefSeq, Oct 2015]

HIST2H2BD: Core component of nucleosome. Nucleosomes wrap and compact DNA into chromatin, limiting DNA accessibility to the cellular machineries which require DNA as a template. Histones thereby play a central role in transcription regulation, DNA repair, DNA replication and chromosomal stability. DNA accessibility is regulated via a complex set of post-translational modifications of histones, also called histone code, and nucleosome remodeling. Belongs to the histone H2B family. The nucleosome is a histone octamer containing two molecules each of H2A, H2B, H3 and H4 assembled in one H3-H4 heterotetramer and two H2A-H2B heterodimers. The octamer wraps approximately 147 bp of DNA
