Testing Data

(HTRF assay for HIPK3 (163-562) activity 1 uM STK S3 substrate was incubated with different concentrations of HIPK3 (163-562) protein in a 10 ul reaction system containing 1×Enzymatic Buffer, 5 mM MgCl2, 1 mM DTT, and 100 ?M ATP for 1 hour. Then 10 ul detection reagents containing STK antibody (1:2) and SA-XL665 (1:100) diluted with 1× Detection Buffer were added and incubated with the reactions for 30 min. All the operations and reactions were performed at room temperature. HTRF assay was used for detection.)

Related Product Information for HIPK3 recombinant protein
Background: HIPK3 (Homeodomain Interacting Protein Kinase 3), also known as FIST. It is a serine/threonine-protein kinase involved in transcription regulation, apoptosis and steroidogenic gene expression, HIPK3 phosphorylates JUN and RUNX2. It seems to negatively regulate apoptosis by promoting FADD phosphorylation and enhances androgen receptormediated transcription. It may also act as a transcriptional co-repressor for NK homeodomain transcription factors. In osteoblasts, HIPK3 supports transcription activation and phosphorylates RUNX2 that synergizes with SPEN/MINT to enhance FGFR2 mediated activation of the osteocalcin FGF-responsive element (OCFRE).

Product Notes

The HIPK3 hipk3 (Catalog #AAA389408) is a Recombinant Protein produced from Baculovirus and is intended for research purposes only. The product is available for immediate purchase. It is sometimes possible for the material contained within the vial of "HIPK3, Recombinant Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
