Applicable Applications for CREB3L1 cell lysate
Western Blot (WB)
Application Notes
WB: Use at an assay dependent dilution.
Species
Human
Recommend Usage
1. Centrifuge the tube for a few seconds and ensure the pellet at the bottom of the tube.2. Re-dissolve the pellet using 200uL pure water and boil for 2-5 min.
Sequence Construction
A DNA sequence encoding the human CREB3L1 (Q96BA8-1) (Glu396-Ser519) was expressed with a polyhistidine tag at the C-terminus.
Preparation Method
Cell lysate was prepared by homogenization of the over-expressed cells in ice-cold modified RIPA Lysis Buffer with cocktail of protease inhibitors. Cell debris was removed by centrifugation. Protein concentration was determined by Bradford assay (protein assay, Microplate Standard assay). The cell lysate was boiled for 5 min in 1 x SDS loading buffer (50mM Tris-HCl pH 6.8, 12.5% glycerol, 1% sodium dodecylsulfate, 0.01% bromophenol blue) containing 5% b-mercaptoethanol, and lyophilized.
Preparation and Storage
Store at 4 degree C for up to twelve months from date of receipt. After re-dissolution, aliquot and store at -80 degree C for up to twelve months. Avoid repeated freeze-thaw cycles.
Samples are stable for up to twelve months from date of receipt.
Related Product Information for CREB3L1 cell lysate
CREB3L1, also known as OASIS, is a cellular transcription factor synthesized as a membrane-bound precursor. It is a putative endoplasmic reticulum (ER) stress sensor in astrocytes with a mechanism of activation. OASIS mRNA expression was detected in pancreatic beta-cell lines and rodent islets, and the expression level was up-regulated by ER stress-inducing compounds. CREB3L1 may have a role in pancreas development. CREB3L1 may also play an important role in limiting virus spread by inhibiting proliferation of virus-infected cells. In vitro, CREB3L1 binds to box-B element, cAMP response element (CRE) and CRE-like sequences, and activates transcription through box-B element but not through CRE. It may play a role in gliosis.

This Human CREB3L1 overexpression lysate was created in HEK293 Cells and intented for use as a Western blot (WB) positive control. Purification of CREB3L1 protein from the overexpression lysate was verified.

NCBI Description

The protein encoded by this gene is normally found in the membrane of the endoplasmic reticulum (ER). However, upon stress to the ER, the encoded protein is cleaved and the released cytoplasmic transcription factor domain translocates to the nucleus. There it activates the transcription of target genes by binding to box-B elements. [provided by RefSeq, Jun 2013]

Uniprot Description

CREB3L1: Transcription factor that acts during endoplasmic reticulum stress by activating unfolded protein response target genes. Specifically involved in ER-stress response in astrocytes in the central nervous system (By similartity). May play a role in gliosis. In vitro, binds to box-B element, cAMP response element (CRE) and CRE-like sequences, and activates transcription through box-B element but not through CRE. Belongs to the bZIP family. ATF subfamily. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: Membrane protein, integral; Transcription factor

Chromosomal Location of Human Ortholog: 11p11.2

Cellular Component: endoplasmic reticulum; endoplasmic reticulum membrane; integral to membrane; nucleus

Molecular Function: chromatin binding; protein binding; sequence-specific DNA binding

Product Notes

The CREB3L1 creb3l1 (Catalog #AAA8114985) is a Cell Lysate produced from HEK293 Cells and is intended for research purposes only. The product is available for immediate purchase. The immunogen sequence is 135aa. AAA Biotech's CREB3L1 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB). WB: Use at an assay dependent dilution. Researchers should empirically determine the suitability of the CREB3L1 creb3l1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. The amino acid sequence is listed below: Glu396-Ser 519. It is sometimes possible for the material contained within the vial of "CREB3L1, Cell Lysate" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
