Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for GZMA has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any GZMA present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for GZMA is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of GZMA bound in the initial step. The color development is stopped and the intensity of the color is measured.

Function: Abundant protease in the cytosolic granules of cytotoxic T-cells and NK-cells which activates caspase-independent cell death with morphological features of apoptosis when delivered into the target cell through the immunological synapse. It cleaves after Lys or Arg. Cleaves APEX1 after 'Lys-31' and destroys its oxidative repair activity. Cleaves the nucleosome assembly protein SET after 'Lys-189', which disrupts its nucleosome assembly activity and allows the SET complex to translocate into the nucleus to nick and degrade the DNA

Catalytic activity: Hydrolysis of proteins, including fibronectin, type IV collagen and nucleolin. Preferential cleavage: -Arg-|-Xaa-, -Lys-|-Xaa- >> -Phe-|-Xaa- in small molecule substrates.
