Principle of the Assay||This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti-Human GUCY2C antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-Human GUCY2C antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. HRP-Streptavidin was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the Human GUCY2C amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of Human GUCY2C can be calculated.

Uniprot Description

GUCY2C: Receptor for the E.coli heat-stable enterotoxin (E.coli enterotoxin markedly stimulates the accumulation of cGMP in mammalian cells expressing GC-C). Also activated by the endogenous peptide guanylin. Defects in GUCY2C are the cause of diarrhea type 6 (DIAR6). A relatively mild, early-onset chronic diarrhea that may be associated with increased susceptibility to inflammatory bowel disease, small-bowel obstruction, and esophagitis. Defects in GUCY2C are a cause of meconium ileus (MECIL). A condition characterized by a intestinal obstruction due to inspissated meconium in the distal ileum and cecum, which develops in utero and presents shortly after birth as a failure to pass meconium. Meconium ileus is a known clinical manifestation of cystic fibrosis. Belongs to the adenylyl cyclase class-4/guanylyl cyclase family.
