INTENDED USE: This GPX1 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Porcine GPX1. This ELISA kit for research use only!

PRINCIPLE OF THE ASSAY: GPX1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-GPX1 antibody and an GPX1-HRP conjugate. The assay sample andbuffer are incubated together with GPX1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the GPX1 concentration since GPX1 from samples and GPX1-HRP conjugate compete for the anti-GPX1 antibody binding site. Since the number of sites is limited, as more sites are occupied by GPX1 from the sample, fewer sites are left to bind GPX1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The GPX1 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a member of the glutathione peroxidase family. Glutathione peroxidase functions in the detoxification of hydrogen peroxide, and is one of the most important antioxidant enzymes in humans. This protein is one of only a few proteins known in higher vertebrates to contain selenocysteine, which occurs at the active site of glutathione peroxidase and is coded by UGA, that normally functions as a translation termination codon. In addition, this protein is characterized in a polyalanine sequence polymorphism in the N-terminal region, which includes three alleles with five, six or seven alanine (ALA) repeats in this sequence. The allele with five ALA repeats is significantly associated with breast cancer risk. Two alternatively spliced transcript variants encoding distinct isoforms have been found for this gene.
