Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for AST has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any AST present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for AST is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of AST bound in the initial step. The color development is stopped and the intensity of the color is measured.

Uniprot Description

Function: Biosynthesis of L-glutamate from L-aspartate or L-cysteine. Important regulator of levels of glutamate, the major excitatory neurotransmitter of the vertebrate central nervous system. Acts as a scavenger of glutamate in brain neuroprotection. The aspartate aminotransferase activity is involved in hepatic glucose synthesis during development and in adipocyte glyceroneogenesis. Using L-cysteine as substrate, regulates levels of mercaptopyruvate, an important source of hydrogen sulfide. Mercaptopyruvate is converted into H2S via the action of 3-mercaptopyruvate sulfurtransferase (3MST). Hydrogen sulfide is an important synaptic modulator and neuroprotectant in the brain

Catalytic activity: L-aspartate + 2-oxoglutarate = oxaloacetate + L-glutamate.

Cofactor: Pyridoxal phosphate.
