Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate GMPS in samples. An antibody specific for GMPS has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyGMPS present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for GMPS is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of GMPS bound in the initial step. The color development is stopped and the intensity of the color is measured.

NCBI Description

In the de novo synthesis of purine nucleotides, IMP is the branch point metabolite at which point the pathway diverges to the synthesis of either guanine or adenine nucleotides. In the guanine nucleotide pathway, there are 2 enzymes involved in converting IMP to GMP, namely IMP dehydrogenase (IMPD1), which catalyzes the oxidation of IMP to XMP, and GMP synthetase, which catalyzes the amination of XMP to GMP. GMPS: Involved in the de novo synthesis of guanine nucleotides which are not only essential for DNA and RNA synthesis, but also provide GTP, which is involved in a number of cellular processes important for cell division. A chromosomal aberration involving GMPS is found in acute myeloid leukemias. Translocation t(3,11)(q25,q23) with MLL.
