Principle of the Assay: GAP43 ELISA kit applies the competitive enzyme immunoassay technique utilizing a Polyclonal anti-GAP43 antibody and an GAP43-HRP conjugate. The assay sample and buffer are incubated together with GAP43-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the GAP43 concentration since GAP43 from samples and GAP43-HRP conjugate compete for the anti-GAP43 antibody binding site. Since the number of sites is limited, as more sites are occupied by GAP43 from the sample, fewer sites are left to bind GAP43-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The GAP43 concentration in each sample is interpolated from this standard curve.
NCBI and Uniprot Product Information
NCBI Description
The protein encoded by this gene has been termed a 'growth' or 'plasticity' protein because it is expressed at high levels in neuronal growth cones during development and axonal regeneration. This protein is considered a crucial component of an effective regenerative response in the nervous system. Alternatively spliced transcript variants encoding distinct isoforms have been found for this gene. [provided by RefSeq, Jul 2008]
Uniprot Description
GAP43: This protein is associated with nerve growth. It is a major component of the motile growth cones that form the tips of elongating axons. Plays a role in axonal and dendritic filopodia induction. Identified in a complex containing FGFR4, NCAM1, CDH2, PLCG1, FRS2, SRC, SHC1, GAP43 and CTTN. Binds calmodulin with a greater affinity in the absence of Ca(2+) than in its presence. Belongs to the neuromodulin family. 2 isoforms of the human protein are produced by alternative splicing.

Product Notes
The Rat GAP43 gap43 (Catalog #AAA723441) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA723441 ELISA Kit recognizes Rat GAP43. It is sometimes possible for the material contained within the vial of "Growth Associated Protein 43, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
