Reactivity
Porcine
Specificity
This assay has high sensitivity and excellent specificity for detection of FBN. No significant cross-reactivity or interference between FBN and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between FBN and all the analogues, therefore, cross reaction may still exist in some cases.
Samples
Serum, plasma, cell culture supernatants, body fluid and tissue homogenate
Assay Type
Quantitative Competitive
Sensitivity
1.0 ng/mL

Intended Uses: This FBN ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Porcine FBN. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: FBN ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-FBN antibody and an FBN-HRP conjugate. The assay sample and buffer are incubated together with FBN-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the FBN concentration since FBN from samples and FBN-HRP conjugate compete for the anti-FBN antibody binding site. Since the number of sites is limited, as more sites are occupied by FBN from the sample, fewer sites are left to bind FBN-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The FBN concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes fibronectin, a glycoprotein present in a soluble dimeric form in plasma, and in a dimeric or multimeric form at the cell surface and in extracellular matrix. Fibronectin is involved in cell adhesion and migration processes including embryogenesis, wound healing, blood coagulation, host defense, and metastasis. The gene has three regions subject to alternative splicing, with the potential to produce 20 different transcript variants. However, the full-length nature of some variants has not been determined. Uniprot Description

FN1: Fibronectins bind cell surfaces and various compounds including collagen, fibrin, heparin, DNA, and actin. Fibronectins are involved in cell adhesion, cell motility, opsonization, wound healing, and maintenance of cell shape. Defects in FN1 are the cause of glomerulopathy with fibronectin deposits type 2 (GFND2); also known as familial glomerular nephritis with fibronectin deposits or fibronectin glomerulopathy. GFND is a genetically heterogeneous autosomal dominant disorder characterized clinically by proteinuria, microscopic hematuria, and hypertension that leads to end-stage renal failure in the second to fifth decade of life. 15 isoforms of the human protein are produced by alternative splicing.

Product Notes

The Porcine FN fn1 (Catalog #AAA741974) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA741974 ELISA Kit recognizes Porcine FN. It is sometimes possible for the material contained within the vial of "Fibronectin, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
