INTENDED USE This FN ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Canine FN. This ELISA kit for research use only!

PRINCIPLE OF THE ASSAY FN ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-FN antibody and an FN-HRP conjugate. The assay sample and buffer are incubated together with FN-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the FN concentration since FN from samples and FN-HRP conjugate compete for the anti-FN antibody binding site. Since the number of sites is limited, as more sites are occupied by FN from the sample, fewer sites are left to bind FN-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The FN concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes fibronectin, a glycoprotein present in a soluble dimeric form in plasma, and in a dimeric or multimeric form at the cell surface and in extracellular matrix. Fibronectin is involved in cell adhesion and migration processes including embryogenesis, wound healing, blood coagulation, host defense, and metastasis. The gene has three regions subject to alternative splicing, with the potential to produce 20 different transcript variants. However, the full-length nature of some variants has not been determined. FN1: Fibronectins bind cell surfaces and various compounds including collagen, fibrin, heparin, DNA, and actin. Fibronectins are involved in cell adhesion, cell motility, opsonization, wound healing, and maintenance of cell shape. Defects in FN1 are the cause of glomerulopathy with fibronectin deposits type 2 (GFND2); also known as familial glomerular nephritis with fibronectin deposits or fibronectin glomerulopathy. GFND is a genetically heterogeneous autosomal dominant disorder characterized clinically by proteinuria, microscopic hematuria, and hypertension that leads to end-stage renal failure in the second to fifth decade of life. 15 isoforms of the human protein are produced by alternative splicing.
