Principle of the assay: FLNa ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-FLNa antibody and an FLNa-HRP conjugate. The assay sample and buffer are incubated together with FLNa-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the FLNa concentration since FLNa from samples and FLNa-HRP conjugate compete for the anti-FLNa antibody binding site. Since the number of sites is limited, as more sites are occupied by FLNa from the sample, fewer sites are left to bind FLNa-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The FLNa concentration in each sample is interpolated from this standard curve.

The protein encoded by this gene is an actin-binding protein that crosslinks actin filaments and links actin filaments to membrane glycoproteins. The encoded protein is involved in remodeling the cytoskeleton to effect changes in cell shape and migration. This protein interacts with integrins, transmembrane receptor complexes, and second messengers. Defects in this gene are a cause of several syndromes, including periventricular nodular heterotopias (PVNH1, PVNH4), otopalatodigital syndromes (OPD1, OPD2), frontometaphyseal dysplasia (FMD), Melnick-Needles syndrome (MNS), and X-linked congenital idiopathic intestinal pseudoobstruction (CIIPX). Two transcript variants encoding different isoforms have been found for this gene.[provided by RefSeq, Mar 2009]

FLNA: a ubiquitous cytoskeletal protein that promotes orthogonal branching of actin filaments and links actin filaments to membrane glycoproteins. Plays an essential role in embryonic cell migration. Anchors various transmembrane proteins to the actin cytoskeleton and serves as a scaffold for a wide range of cytoplasmic signaling proteins. Interactions with filamin A may allow neuroblast migration from the ventricular zone into the cortical plate. Tethers cell surface-localized furin, modulates its rate of internalization and directs its intracellular trafficking.
